Thymosin beta4 promotes cardiac repair following myocardial infarction in the rat model[1]

This study examined thymosin beta-4 (TB-500) following surgically induced myocardial infarction in adult rats to evaluate its effect on cardiac repair and function in a preclinical model. TB-4 was injected intraperitoneally beginning immediately after infarction and for seven subsequent days. Echocardiographic measurements at four weeks demonstrated significantly preserved left ventricular ejection fraction and reduced infarct size in research subjects. Histological analysis revealed enhanced angiogenesis and reduced fibrosis in the infarct border zone, consistent with TB-4's known role in actin dynamics and endothelial cell migration.

The anti-inflammatory peptide Ac-SDKP is released from thymosin beta-4 by renal meprin and prolyl oligopeptidase[2]

This biochemical study characterized the enzymatic release of the anti-inflammatory tetrapeptide Ac-SDKP from its precursor thymosin beta-4 in kidney and plasma. Using purified renal meprin and prolyl oligopeptidase, researchers demonstrated that both enzymes cleave the N-terminal Ac-SDKP sequence from TB-4 under physiological conditions. Ac-SDKP inhibited NF-kB-dependent pro-inflammatory gene expression in renal tubular epithelial cells in a concentration-dependent manner. These findings established the molecular basis for TB-4's anti-fibrotic and anti-inflammatory bioactivity through its cleavage product rather than the intact peptide.
